Keep calm and listen to the squid: from fertilization to CRISPR-Cas 9 genome editing with Doryteuthis (Loligo) pealeii

Deciphering the patterns that emerge during development and regeneration drive the research questions in the Crawford laboratory. The systems explored include regeneration and development in the Mexican axolotl, regeneration in the California Blackworm, and development in The Atlantic longfin squid. While Dr. Crawford's academic career has been based at Maryland’s public liberal arts honors college, curiosity and serendipity has taken her back many summers to the Marine Biological Laboratory in Woods Hole, MA, where she attended the Embryology Course as a graduate student. At that time, while enchanted by every new embryo, it was the cephalopod that captured her imagination. In this presentation, Dr. Crawford will describe establishing in vitro fertilization and embryo culture methods to explore early development in the squid embryo and share her journey to create injection methods that enable us to analyze cell lineage relationships and employ CRISPR-Cas 9 genome editing to explore conserved patterns of gene expression during development within this group.
